The Shrimpers were quickly brought back down to earth after their 5-3 penalty shootout win over Colchester on Wednesday.
Their return to League Two action saw them caned 4-0 by Darlo, who were 3-0 up at half-time.
Alun Armstrong gave the hosts the lead after 15 minutes and then Clyde Wijnhard doubled the advantage nine minutes later with his debut Darlington goal.
Armstrong made it 3-0 on 40 minutes and seven minutes after the restart he completed his hat-trick for the Quakers.
