Goals from Leon Constantine and Steven Clark plus a brace from Drewe Broughton saw the Seasiders set up an intriguing two-legged tussle with the Layer Road side, who won their semi final 3-2 at Northampton Town.
"It will be great," said Tilson. "It's a local derby and I'm sure both sets of fans are looking forward to it already.
"I haven't seen Colchester that much this season, but there will be two good crowds for what should be two good games.
"Both sides also have the huge incentive of being so close to playing at the Millennium Stadium."
Tilson admitted he was delighted with his side's impressive display against promotion-pushing QPR.
"They haven't let many goals in this season and they're the in form team in Division Two at the moment so it's a great result," said the 37-year-old.
"They're flying high but in the end we were quite comfortable."
