Benfleet lost by seven wickets as the Old Boys won with just three balls to spare.
Jonathan Edwards hit a healthy 81 to lead the way after Benfleet won the toss and decided to bat.
Edwards was supported by Ben West (34), skipper Mike Tremain (29) and an unbeaten 26 from David Wright as the visitors made 222-9 off their 52 overs.
Spinner Tom Robson took 3-54 for Old Brentwoods, who found their winning reply largely through Roy (102) and skipper Buttleman (82).
Benfleet bowlers Ben West (1-72) and Stephen Scott (2-65) led the attack well but received scant reward for their efforts.
"All credit to the way Ben and Stephen set about it," said Benfleet spokesman Ron Steed.
"They bowled superbly but it was one of those things The bowlers put the ball in the right places but they got hit."
